MULTISCREEN™ division-arrested cells are derived from our high-quality MULTISCREEN™ GPCR stable cell lines. The target can be chosen from an extensive catalog of 500+ GPCR stable cell lines including human and ortholog, such as mouse, rat, rabbit, dog, and monkey.  The majority of these human and ortholog GPCR target pairs share matching pharmacology and expression. Moreover, division-arrested parental cells for each target are available and recommended as negative controls.

MULTISCREEN™ GPCR division-arrested cells are developed for one 384- or 96-well microplate-based assays. Therefore, they are low-cost for evaluating a small number of compounds. They may be used in counter-screens for ortholog targets, closely related GPCRs that may cross-react to the same compounds, or just another assay for signaling pathway bias. They can also be used to test tool compounds in pilot projects before committing to purchasing MULTISCREEN™ GPCR stable cell lines. In addition to 4 million-cell cryovials, MULTISCREEN™ GPCR division-arrested cells are also available at a higher density upon request (billions of cells in bulk aliquots for HTS).

How well do they perform?

To ensure such high quality performance, we aim to obtain pharmacological profiles in our division-arrested cells similar to those in growing stable cell lines.

Each MULTISCREEN™ GPCR stable cell line is rigorously validated for assay function, clonality, and stability. In addition, Multispan’s division-arrested cells also maintain the multi-screen nature of multiple assays in each MULTISCREEN™ stable cell line, or ALL-IN-ONE for all relevant assays in one single cell line. We bank and validate each new batch of assay-ready cells based on a specific assay requested. Frequently they are validated in the primary GPCR signaling assays including the calcium assay, cAMP assay, and beta-arrestin assay. They are also at times validated in the binding assay by flow cytometry (FACS) for the purpose of screens that are focused on receptor binding. 

Moreover, to preserve the high performance of our stable cell lines and batch-to-batch consistency, Multispan’s scientists have developed extensive proprietary processes in cell culture scaling up, growth-arrest, freezing and thawing. Each batch of MULTISCREEN™ assay-ready cells is QC’ed in a functional assay side-by-side with its original stable cell line as an additional level of quality assurance.
