MULTISCREEN™ TR-FRET cAMP 1.0 No-Wash Assay Kits provide a homogeneous assay method for adenylyl cyclase activity detection in 96-well, 384-well, and higher multiplexity. The dual-label system uses an α-cAMP antibody labeled with MULTISCREEN™ Eu and cAMP labeled with MULTISCREEN™ 650.

In the absence of free cAMP, MULTISCREEN™ 650-cAMP binds to MULTISCREEN™ Eu-α-cAMP-Ab, producing a strong TR-FRET emission at 655 nm. Free cAMP in the sample competes for binding to the antibody, reducing the TR-FRET signal. Because MULTISCREEN™ 650-cAMP has a fluorescence lifetime of only nanoseconds while MULTISCREEN™ Eu-α-cAMP-Ab has a much longer lifetime, the distinct fluorescence lifetimes enable precise determination of cAMP concentration, with the TR-FRET magnitude proportional to the cAMP in the sample.
Our Kits our compatible with most commercially available multimode microplate readers, and are validated with MULTISCREEN™ stable cell lines.
cAMP Dose-Response in MULTISCREEN™ β1-HEK293T stable cell line (C1437-1) measured with MULTISCREEN™ TR-FRET cAMP 1.0 No Wash Assay Kit. Cells were seeded in a 384-well solid white plate, incubated with Isoproterenol and 1mM IBMX before being treated with MULTISCREEN™ Eu-α-cAMP-Ab and MULTISCREEN™ 650-cAMP, and measured by FlexStation 3.
